The aim of this project is to transfer the ToF-SIMS technique expertise gained in the project GR/N20546 (developing a 'chemical microscope' for biological research) to a laboratory deeply involved in metabolomics research. Present investigations have showed that ToF-SIMS analysis is capable of visualising biochemicals from a variety of cellular systems including freeze-fractured yeast cells. This study is undertaken to develop ToF-SIMS capability for metabolic engineering using Saccharomyces cerevisiae as a model system. The proposed study aims at identifying metabolic intermediates in biochemical pathways. We will determine the extend to which ToF-SIMS can identify specific metabolites, initially in the culture liquid and ultimately spatially resolved within individual yeast cells. Extending these metabolite studies to yeast mutants could give further insights into intracellular metabolism and its regulation. Success in this pilot study will pave the way for the use of ToF-SIMS in two important areas of metabolomics research. Firstly as a high throughput technique for the direct analysis of multiple samples in an array, with little or no sample preparation. Secondly to detect changes in the metabolome profile directly in situ.
