Hypothesis: A colloidal carrier for the delivery of polypeptides by the pulmonary route will enhance drug bioavailability if it rapidly spreads on deposition in the airways. This proposal is to examine a number of lipid- based colloidal carriers and seek to improve their dispersion into a spread lipid monolayer by utilising the endogenous apoproteins SP-B and SP-C. The rate and extent of incorporation will be determined using a NIMA Langmuir Blodgett trough, as monolayer penetration experiments are a valuable approach to determining interactions between lipid monolayers and polypeptides. As surface viscosity is important for lung surfactant function, we propose to employ an oscillating interfacial rheometer to investigate the rheological changes (interfacial shear elasticity and viscosity) resulting from polypeptide and apoprotein dispersion into the spread monolayers. The thermotropic behaviour of peptide-surfactant mixtures will be examined by FTIR; lipid- peptide monolayers transferred to ATR crystals should enable interpretation of peptide interaction based on induced changes in the phospholipid acyl chain configuration. High sensitivity DSC will be used to probe the influence of the apoproteins and polypeptides on the thermal transitions of the lipidic carriers.
