This proposal is for equipment to underpin collaborative and multidisciplinary research on self-assembling systems of pharmaceutical relevance. Laser light scattering will be utilised in the characterisation of the self-association in aqueous solution of (a) block copolymers designed to enhance the solubilisation and delivery of poorly soluble drugs and (b)selected amphiphilic drugs. The micelles of block copolymers have potential for the solubilisation and delivery of drug molecules, but their exploitation for this purpose is limited by their low solubilisation capacity and their instability in the blood stream. To enhance the solubilisation capacity, novel block copolymers have been designed that form micelles with cores that are aromatic or are of variable hydrophobicity in order to provide favourable sites for drug solubilisation. Stable micelles produced by polymerisation of functional end-groups of block copolymers within the micelle cores will also be examined. The self-association of surface active drugs may have important consequences for their biological activity. It is proposed to examine the association characteristics of selected drugs that show potential for association or which form a structurally related series of use for probing links between molecular structure and association pattern.
