It is proposed to continue with a research programme concerned with completing a total synthesis of bryostatins. A new approach recently developed for the synthesis of cis-2,6-6-disubstituted tetrahydropyrans is to be applied to complete a synthesis of the C(5) -C(16) fragment of the bryostatins and the product obtained will be elaborated to complete a synthesis of the C(1) - C(16) fragments. This will then be coupled to the C(17) - C(27) fragment using a Julia reaction and the product taken through to complete a synthesis of bryostatin 11. Preliminary studies of a synthesis of bryostatin 3 will also be undertaken.
