Integral to the development and assessment of lower limb prostheses is improved understanding of the role played by wear debris in osteolysis, the most common cause of aseptic loosening of total hip replacements (THR). Comprehensive characterisation wear particles produced by a replacement joint is pivotal to much of the research in this area. The proposers have developed a novel, non destructive debris characterisation approach which yields detailed and accurate quantification of both ex vivo and in vitro origin wear debris, obviating the need for sequential filtration, weighing and SEM 2-D size measurement. Funds are sought to investigate ex vivo wear debris from aseptic failed THRs. The project will investigate the size distribution, the morphology, the mobility and the material properties of wear particles generated by clinically failed THRs, combine this with collaborative clinical and histological assessments of osteolysis and so quantify the role of wear debris in joint failure. Wear debris produced in vitro via hip simulator testing will then be characterised and compared with that retrieved ex vivo to enable the refinement of hip simulator loading and motion conditions to simulate the in vivo situation more accurately.
