Stereopure alpha-C-substituted alpha-amino acids are an important class of compounds. Simple representatives and their derivatives are already in the market place. More complex representatives are constituents of numerous natural products that are of therapeutic relevance. Peptides in which alpha-amino acids are replaced by alpha-alkyl alpha-amino acids are of growing note because of their restricted conformational properties, enabling folding and molecular recognition phenomena to be defined. One of the most powerful methods for the synthesis of alpha-alkylated alpha-amino acids uses readily available alpha-amino acids as starting materials. It requires the installation of a new temporary stereogenic centre to direct the alkylation reaction; the temporary centre is then destroyed. In principle, it should be possible to do away with these two steps, and therefore streamline the synthesis, by imprinting a stereochemical memory on the intermediate during its generation. The basis of this proposal is to understand the rules that will lead to successful memory imprints and to apply them to the synthesis of stereopure alpha-C-substituted alpha-amino acids.
