The proposed project involves culturing human dermal fibroblasts and seeding them in a standard rectangular collagen gel with fibroncetin stands perpendicular to the long axis. Mechanical load will then be applied to the gels using a locally produced instrument the tensioning-culture force monitor along the long axis of the gel causing cell alignment in tow directions. The load applied will be static and cyclic(1-5 cycles/hour). The effects on the production of Collagen Type I,III,MMP'sl,2,3,8,9,TIMP'sl 1 and 2 and growth quantitative RT-PCR using specific primers and templates. This proposed study will help in the understanding of cellular and matrix organisation in the tissue engineering on biomaterials and define the effects of dual cues (mechanical and contact guidance) on matrix production.
