New conducting polymer/bioreagent film conbinations will be produced, utilising alkyl and coarboxylic acid derivatives of pyrrole and thiophene, to enable direct reagentless sensing of affinity molecules binding in the absence of a reaction product or indicator reagent label. Key to this will be the identification of impedence behaviour of supporting conductive polymer films as the resuls of analyte bimolecular interactions. The analytical targets will be the reproductive hormone, LH, FSH, and a range of drugs with polar/hydrophobic properties of high clinical relevance which will also serve as model systems. As a platform for this work, a range of model enzymes, antibodies and lectins, will be studied and as appropriate developed to a stage of biofluid analysis. Electrochemical and optical interrogation techniques will be used to gain insight into the transduction mechanisms. As part of this work, the kinetics of analyte interaction will be followed in real time and realted to film thickness, biomolecule loading and electrode design. Effects of measurement conditions for the various conducting polymer/bioaffinity molecule constructs will be determines. The incorporation of membrame barriers will be a key part in the project, enabling direct measurments of analytes in serum, blood and urine samples.
