The aim of this project is to investigate the feasibility of using solution calorimetry to predict oral drug bioavailability and measure the effects of absorption-enhancing lipid-based formulations. Calorimetry has the unique ability to make quantitative in situ measurements of the processes that occur in simulated fed or fasted state intestinal fluid after the administration of a drug in a digestible lipid delivery vehicle. The heat of solution of model compounds in partially digested lipid formulations in simulated intestinal fluids will be compared to in vitro drug permeability data obtained using the same systems.
