Three-membered carbon ring systems, known as cyclopropanes, have a highly strained, rigid structure, which can prove useful in designing molecules with desirable properties, eg in drug design, correctly positioning atoms to interact with an enzyme. At present, the methods available to organic chemists to make these three-membered rings are limited and generally involve the use of very toxic chemicals as reagents and starting materials which are extrememly hazardous to handle, particularly on large scale. Only a very limited range of these starting materials can be bought from commercial suppliers, and the preparation of more complicated examples is both difficult and potentially dangerous.Over the past twenty years or so, our group has developed new methods for making these structures, that make use of considerably milder chemicals as starting materials. As well as being considerably easier to handle, an enormous range of these compounds can be easily bought and non-commercially available derivatives are easily prepared using generally non-hazardous chemical processes. In this proposal, we hope to extend the methods we have developed to the construction of more complex cyclopropanes, which will be of direct use in the future development of molecules with useful medicinal and chemical properties.
