Many natural products are of physiological interest - but their properties are only exploitable if they can be extracted from natural sources sufficiently straightforwardly that large quantities are available. Members of the kainoid family of neurotransmitter analogues are immensely valuable in neurophysiological research, allowing the processes associated with nerve cell death to be studied. However, few are readily available from natural sources and even naturally extracted kainic acid has become scarce. We propose to develop a practical synthesis of domic and the isodomic acids - potent relatives of kainic acid most of which have never been synthesised and which are currently unavailable from natural sources. We shall use our dearomatising anionic cyclisation reaction to convert cheap aromatic starting materials into two key intermediates by efficient fragmentation reactions. Our versatile route will lead to almost all known members of the domoic family by using Pd-catalysed coupling chemistry to add their respective side-chains at a late stage in the synthesis.
