|
The high sensitivity of surface enhanced Raman scattering (SERS) and the incisiveness of Raman optical activity (ROA) will be combined to produce a new spectroscopic technique called surface enhanced Raman optical activity (SEROA). A combined Raman/ROA spectrometer will be built and used to collect SERS and SEROA spectra of ribonucleosides and ribonucleotides adsorbed onto the surfaces of silver and gold colloids. Optimum conditions of colloid surface morphology, size and activation for SEROA will be established and the SEROA spectra will be analyzed using two-dimensional correlations with complementary SERS, Raman and ROA data. The resultant band assignments will be used to investigate the interactions between the different adsorbates and colloid surfaces with the aim of characterizing the modes of adsorbate coordination (through purine/pyrimidine rings, ribose hydroxyls or phosphate groups) and adsorbate stereochemistry. The spectra of related adsorbates will also be analyzed to determine if SEROA can be used as a tool for structural fingerprinting of molecules.
