During the circulation of the blood fluid components are filtered out of the blood vessels into surrounding tissues. This fluid is returned to the general blood circulation via a network of vessels known as the lymph system. Failure of the lymph system due to disease or surgery can have disastrous consequences, including tissue swelling, impaired functioning of organs and breakdown of the immune system. Lymph flow is driven by the phasic contraction of segments of the vessel wall which are separated by passive, uni-directional valves. We have developed in vitro preparations in which we have extensively studied the physiology of the lymphatic vessels. We now propose to extend this study with measurements of the essential fluid mechanical parameters such as local pressures, wall geometry and mechanical properties, flow visualisation and computational modelling to investigate flow in individual lymph vessels and in lymph networks. In this, the first study to link all of these techniques, we shall establish the influence of a range of physiological parameters on pumping efficiency. The data obtained will be of value in understanding many medical conditions.
