The proposed research programme involves the first ever full and systematic study into the use of deuterium for the protection of carbon-hydrogen bonds (via exploitation of the primary kinetic isotope effect) in important synthetic procedures that involve radical and carbene intermediates. Despite the use of such isotope effects in the study of reaction mechanisms, remarkably few synthetic procedures have ever exploited this principle. After carrying out systematic studies of the fundamental aspects of this methodology, extensions of the work into generally applicable syntheses of a variety of biologically important, highly substituted amines and amino acids are planned. These will allow access to compounds which would be difficult to obtain using more traditional synthetic methods, including peptides containing noncoded amino acids which are generated from simple amino acids already present in a peptide chain. All of the synthetic procedures allow full recovery and recycling of the deuterium protecting group, a point which is generally overlooked in protecting group chemistry.The proposal is a substantially rewritten resubmission of GR/R71313/01 taking into account the comments of the original referees and panel.
