We propose to embark on a programme of synthetic chemistry that builds on preliminary investigations within our group aimed towards the preparation of new organofluorine compounds. The introduction of fluorine atoms in molecules strongly modifies their physical and biological properties leading therefore to useful drugs or interesting pharmacological tools. The regio- and stereocontrol in the fluorination of poorly activated substrates is an unexplored area of research. A novel methodology will be developed and optimised for the preparation of a wide range of functionalised allylic fluorides. The applicability of the methodology to the preparation of several biologically interesting fluorinated targets will be investigated in detail. In the second part of this project, the viability of a novel approach for asymmetric allylic fluorination will be studied. This approach is based on the use of homochiral Nfluoroammonium salts prepared from cinchona alkaloids that have already been used successfully for the preparation of enantiomerically enriched fluoroketone derivatives.
