Atomic force microscopy allows the measurement of intermolecular forces by bringing a probe into contact with a surface and then carefully measuring the force required to remove the probe from the surface, termed the pull-off force. It is also possible to combine the detection of intermolecular interactions with topographical imaging, a technique termed affinity imaging. Recent work at Lancaster has involved the development of a dedicated affinity imaging system for measuring protein interactions with biomaterials. We propose to use this system, developed in the Physics Department and adapt it to an application in the Biology Department. We will use it to map antibody-antigen interactions on tissue sections. We have some preliminary data showing this is feasible and we believe that it offers numerous advantages over conventional immunolabelling systems. Antibody detection systems, such as immunoassay and immunolabelling techniques, are used extensively in fundamental biomedical research and in diagnostic medicine. We believe that our system can improve the speed, spatial resolution and sensitivity of immunolabelling of tissue sections and also allow several antigens to be mapped on a single tissue section. Such a system would offer enormous benefits to the biological and biomedical community.
