The proposed research project will concern the synthesis of hemibrevetoxin B, the smallest member of the brevetoxin family. Although hemibrevetoxin B is significantly smaller than the other brevetoxins, it is a formidable synthetic target possessing four fused cyclic ethers and 10 stereogenic centres. In addition, it presents many of synthetic challenges found in larger congeners. Hemibrevetoxin B is a popular synthetic target but only a small number of rather lengthy total syntheses have been reported and a short and efficient synthesis has yet to be completed.This proposal outlines our plans to explore both linear and two-directional strategies for the synthesis hemibrevetoxin B using our recently developed ring-closing metathesis methodology for rapid ring construction. Both approaches should deliver hemibrevetoxin B with a longest linear sequence of less than 30 steps from readily available starting materials. If time permits, a third convergent approach based on fragment coupling will be explored.
