The synthesis and physicochemical characterisation of a series of well-defined gadolinium complexes, and conjugates possessing one or two innersphere waters that undergo fast exchange under ambient conditions. The elaboration of these systems with 3 or 4 biocompatible, hydrophilic substituents which enhance relaxivity but do not impair water exchange kinetics and may allow reversible binding to serum albumin in order to optimise the overall relaxivity. Using this knowledge, a folate-targeted conjugate will be evaluated based on a high relaxivity Gd complex devised herein, regioselectively coupled to the X-C02H group of folic acid.
