The specific inhibition of glycosyltransferases provides valuable mechanistic information about the mode of action of these poorly understood enzymes and is a potential therapeutic strategy in the treatment of disease. The structures and syntheses of several tricomponent polyhydroxylated nitrogen hetereocycles as transition-state analogue inhibitors of the action of rhamnosyltransferases are proposed. Such inihibitors represent potential agents for the treatment of TB. As a consequence of its blockwise nature this methodology is highly flexible and therefore is also applicable to the synthesis of inhibitors of other glycosyltransferases that are mediators in the mechanisms of disease.This 3-month proposal completes a previous FastTrack grant that was cut short by the successful employment of the PDRA concerned.
