Modified nucleic acids and oligonucleotide mimics with improved affinity and specificity for DNA and RNA, show significant promise as therapeutic or diagnostic agents and as bio-analytical tools. We will develop synthetic methods for the solid phase synthesis of de novo zwitterionic pyrrolidine-phosphonate oligonucleotidemimics (zPOM) and investigate the recognition of DNA and RNA using a variety of physical techniques. zPOM are unlike any other mimics developed to date and possess electronic and conformational properties which make them ideal systems for recognition of natural nucleic acids. Underpinning chemistry of this nature is particularly important in exploiting recent advances in genomics.
