The ability to examine the interfacial reactions of biological and chemical processes with fine lateral resolution in realtime has far reaching Implications in cell and tissue engineering and the pharmaceutical sciences. Cell surface interactions are of extreme importance in cell migration, motility and cell surface coupling. Detailed studies of these parameters are difficult, but gaining an understanding of the quantitative spatio/temporal interactions between cells and culture substrates will allow the development of systems for accelerated wound repair and will accelerate progress in the development of functional electrophysiological bioassay systems.In this project we will demonstrate and explore new techniques in surface plasmon microscopy capable of giving a step change in the achievable lateral resolution. The techniques considered will be widefield, so that they can be implemented as adaptations of a conventional microscope and provide parallel measurement channels. The systems will then be used in two distinct and very important application areas: (i) The development of a functional pharmacalogical bioassay system consisting of electrogenic cells cultured over microfabricated extracellular recording and (ii) studies on cell surface interactions in haptotaxic cell migration in wound healing.
