The plakorsins, untenone A and the manzamenones are a varied array of bioactive, fatty acid derived natural products which have all been isolated from plakortis sponges. It is our proposal, that these compounds are all derived from a common biosynthetic precursor, the highly cytotoxic peroxy ketal, chondrillin, which was first isolated from a chrondrilla sponge in 1976 and subsequently from a plakortis sponge in 1990. A conceptually simple series of transformations, which exploit the inherent reactivity of chrondrillin, are proposed to provide a biosynthetic link between this intriguing natural product and the other fatty acid derivatives. The ultimate aim of our proposed research is to prepare all of the natural fatty acid derivatives, as well as a range of unnatural analogues, in a clean and efficient manner from chondrillin using an approach modeled on our biogenetic theory. The brevity of our proposed route will allow the preparation of significant quantities of both natural and unnatural materials which will be assessed for their toxicity towards a range of cancer cell lines.
