A single ion channel is usually modelled by a finite state space continuous time Markov, chain. The state space is partitioned into open and closed states and it is possible to observe only which class of states the channel is in. Short sojourns of the channel in either the open or closed classes of states fail to be detected. This phenomenon, which is known as the time interval omission, hampers inferences concerning ion channel gating mechanisms from observed single channel recordings.In many single channel experiments, it has been observed that channel openings occur in bursts. The aim of the proposed research is to develop inferential methods for ion channel data which are based on properties of bursts and are robust to time interval omission. Specifically, (i) pseudo-likelihood-based estimates of single-channel gating parameters from suitably defined functions (such as the total charge transfer) of successive bursts will be developed and their properties investigated; and (ii) structural inferences concerning a single channel gating mechanism, based on probability density functions and correlation functions of total charge transfer and other properties of bursts, will be explored. Successful completion of the proposed research will provide new ways of overcoming some of the problems caused by time interval omission in single channel analysis.
