We propose to design, set up and calibrate a FRET (Fluorescence Resonance Energy Transfer) microscope which will be optimised for the study of protein complexes in living cells. The system will be used to elucidate the role of the mitotic cyclin-cdk (cyclin dependent kinase) complexes during mitosis of mammalian cells. In-vivo FRET will allow us to establish when and where the complex is formed and proteolysed during the cell cycle. As the major regulators of cell growth, DNA replication, and cell division, cyclin-cdk complexes are widely implicated in a large number of proliferative diseases. The proposed research would allow us, for the first time, to elucidate their function in-situ, under physiologically relevant conditions. Previous studies suggest that the cyclin-cdk complex should be well suited for the application of FRET and thus the proposed research also serves as a validation of FRET as a tool to investigate protein function in living cells.
