This proposal describes the preparation and application to diastereoselective synthesis of enantiopure atropisomeric bisquinazolineones. There are three stages to the work. First, to extend the range of bisquinazolineones which have been synthesised and deracemised to include those bearing a wide range of functional groups including prochiral unsaturation and nucleophilic heteroatoms. This builds on preliminary results which indicate that the synthesis of such compounds should be feasible and that they may be deracemised by a crystallisation-induced asymmetric transformation.The second goal is to use the chiral axis of bisquinazolineones to induce asymmetry in reactions of the substituents. Preliminary results have indicated that epoxidation of prochiral double bonds on the bisquinazolinone scaffold proceed with high levels of diastereoselectivity. Optimisation of these levels, and extension to furthur examples will be pursued. Other reactions such as cycloadditions and the Pauson Khand cyclisation will be investigated. The use of biQs as chiral reagents in asymmetric aldol chemistry will be investigated and optimised.The cleavage of enantio-enriched products from the biquinazolinone scaffold will be optimised. Acid hydrolysis has been shown to to yield carboxylic acids and the prochiral scaffold (bisanthranoly hydrazine) which will be recycled. Milder conditions, more compatible with non-racemisation of centres will be sought.
