This proposal describes the development of a new strategy for the use of a chiral auxiliary for the asymmetric synthesis of chiral aldehydes. The strategy employed for chiral auxiliary design relies on a stereoselective aldol reaction to generate an aldolate fragment that contains a 'temporary' stereogenic hydroxyl group. This hydroxyl group is then employed to direct areange of stereoselective reactions within the aldolate side-chain, to creat one or more new stereogenic centres. Finally, the 'temporary' hydroxyl stereogenic centre is destroyed, by virtue of a retro-aldol reaction, to regenerate the chiral auxiliary fragment and liberate the desired enantiopure aldehyde fragment. It is proposed that this approach is ideally suited to the iterative combinatorial synthesis of highly functionalised natural product fragments that contain side-chains comprosed of a series of repeating sub-units of defined stereochemistry. A strategy for the transferral of this methodology to polymer support for the combinatorial synthesis of libraries of enantiopure aldehydes is also described.
