The final objective of this work is to create a lipid bilayer membrane on a gold surface with properties identical to biological membranes found both on the outside and within cells. Such a system should then allow the incorporation of complex membrane proteins, whose role is vital to life, but are poorly understood when studied in 'real' biological systems. This has applications in many current areas of interest including drug discovery and clinical biological sensing.In order to create such solid supported bilayers it is necessary to 'build up' a molecular support architecture. An understanding of the molecular design of the support layer, and the behaviour of lipids molecules on such supports is critical to the construction of reliable supported lipid bilayer systems. However, many of the factors are poorly understood. This project seeks to first increase our understanding of the basic chemistry and physics involved in the formation of solid-supported membranes and then to construct reliable systems in which membrane proteins will be studied.
