An instrument will be developed that combines a novel, rapid fluorescence imaging microscope with the simultaneous collection and storage of electrica signals from biological cells. The microscope will be designed around a novel light source, based on InGeN LEDs, and will include a method of obtainirn optical sections without the requirement to move the sample or objective lens in the z-plane. It wall have the ability to record images with sub-micron resolution at up to 100fps. The optics will be optimised for imaging fluorescent indicators that specifically report the calcium ion concentration within a cell. New software will be developed to enable the integrated, collection, storage and analysis of time-linked image and electrical signals, so that image and electrical signals can be recorded simultaneously from tlleaame'cel1. The neVvi'jh$trument will provide biological researchers with the ability to visualise the localisation of calcium ions within the cell, which are either the cause or effect of simultaneous electrical signals that arise in the cell membrane due to the activity of membrane ionic channels. .T'he microscope will have the additional capability of recording an infrared transmission image, which will place the calcium signals in theirlocation within the cell.:
