This project seeks to exploit the unique capabilities of tandem ion trap/ion mobility spectrometry, to probe conformational changes resulting from non-covalent ligand-receptor interactions of biomolecules and to relate these to their gas and solution phase structures. In tandem ion trap/ion mobility spectrometry, the ion trap may be operated as a conventional mass analyser or as an ion source for the ion mobility spectrometer. For ion mobility measurements, pulses of ions mass-selected in the ion trap are transferred to a drift tube maintained at a constant pressure of helium (-1-6 Ton-). The mobility of an ion is given by the time taken for the ion to traverse the drift tube under the influence of an electric field gradient and is determined by the ion coMsion cross section (i.e. the structure and conformation of the ion). Tandem ion trap/ion mobility measurements, supported by molecular modelling studies, will be directed towards elucidating the gas and solution phase conformations of ligand-receptor complexes of amino acid and peptide ligarxis with polyether and with glycapeptide antibiotic receptors.
