Traditional oligosaccharide synthesis is hampered by the need for moisture sensitive reactants and reagents, coupled with laborious rounds of chromatographic purification. We have shown that iodine vapour can be used to active thioglycosides deposited on tlc plates (i.e. the absence of solvent). In the presence of sugar alcohols, disaccharides can be formed and subsequently separated in situ without work-up. In principle, trisaccharides could be synthesised by exploiting a second dimension of chromatography. Low-tech micro-scale synthetic chemistry on a tlc plate offers substantial scope for multiple parallel synthesis of glycoside libraries, but also suggests scope for scale-up. The aim of this project is to provide proof of concept.
