New and efficient methods for the resolution of simple carbonyl containing molecules are being sought. We have discovered that an acetoacetate proton donor is a reliable method for direct C-protonation of enolates in the presence residual amines. This direct C-protonation using carbonyl chelating proton donors is a new and fundamental discovery. The practical implications are also considerable, exploitable and clearly solves existing problems associated with previous methodology within this area, such as proton return and the use of more specialised 'base-free' enolates. This will allow an increase in substrate diversity.Consequently, the work has direct implications for the synthesis of biomolecules, including catalysis, new cleaner processes, all main themes supported by the EPSRC.
