The unusual structure of the cornexistins coupled with their potent and selective herbicidal activity makes them highly attractive targets for total synthesis. They present a variety of synthetic challenges, especially wuth regards to medium-ring construction and stereocontrol. The objective of the work outlined in this proposal is the development of an efficient and completely general strategy for the synthesis of hydroxycornexistin and cornexistin analogues.An efficient total synthesis of hydroxycornexistin will require the development of new strategies for the construction of medium -ring carbocycles. Two approaches to construction of core of the natural product will be explored. In the first approach, separate ring-closing metathesis reactions will be used to assemble the nine-membered ring and to control the stereochemistry of exocyclic alkene. The second more convergent strategy will employ a domino ring-closing, ring -opening, ring-closing sequence to address the issUues of medium-ring construction and control of the exocyclic alkene geometry in a single operation.
