The objective of this research is the development of a novel strategy for the synthesis of vitamin D analogues. Such compounds, classically known for their role in regulating calcium metabolism have recently been shown to be relevant to the treatment of cancers, hyperparathyroidism, psoriasis etc. 1a-Hydroxyergocalciferol will be sequentially reacted with alkenyl (dimethyl)silyl chlorides and ruthenium or molybdenum carbenes to produce the corresponding cyclic unsaturated silyl ethers, products of ring closing metatheses. Subsequent deletion of the silyl ether tether by desilylation, oxidation, or palladium (O) catalysed coupling should provide novel vitamin D analogues such as 19-hydroxymethyl -1a-hydroxyergocalciferol.
