The palmarumycins, diepoxins, preussomerines and the spiroxins are structurally remarkable natural products isolated from various microbial fermentation broths. These secondary metabolites show diverse biological effects including selective antifungal and antibacterial activities. This proposal concerns an extension of preliminary studies on the total syntheses of these important natural products to further probe reactivity in the compact polyfunctional core and to prepare additional compounds for bioassay as anti-infective agents. Specifically, the total synthesis of enantiomerically pure preussomerines A, B, G and I and several enantiomerically pure diepoxins will be undertaken. These ventures will underscore the use of regioselective and enantioselective oxidation reactions to enhance the complexity of simple spiroketal precursors. Novel double spiroketalisation reactions will be examined in a quest for superior routes to the preussomerines. This work is relevant to the solution of a major biomedical need: the discovery of an effective non-toxic fungicidal agent for use in human patients. At the same time the research should lead to an understanding of the properties of structurally unusual class of spiroketal natural products. The research therefore has potentially significant economic and social benefits.
