Molecular tools have been used to show that foaming in activated sludge is caused by the number of pleiomorphic mycolic acid containing bacteria exceeding a specific threshold. The extension of this work is severely impeded by the cost, difficulty and time consuming nature of quantitative fluorescent in situ hybridisation. As a prelude to a wide ranging study of the causes of foam in UK activated sludge plants we wished to evaluate and calibrate molecular based methods for the quantification of mycolata in the environment. We wish to develop and compare: automated counting of FISH images; slot blot hybridisation; the enzyme amplified sandwich hybridisation assay and measurement of characteristic cell wall components.If we are successful we may be able to make quantitative molecular technology for the examination of foaming organisms available to the water industry and non specialist research laboratories.EngProg\
