The proposed investigation seeks to develop novel methodology for glycosidic bong formation using a latent/active strategy. In particular this work will seek to address the limitations of current methods by improving both donor reactivity and ease of latent-active functional group interconversion. This will have the effect of improving yields while simplifying the process of carbohydrate syntheses. The work will be carried out in three parts (a) a systematic and extensive study of the synthesis of the latent ester moiety, conversion to the active thionoester and subsequent glycoside bond forming reaction, (b) the development of protective group strategies, complementary to the latent anomeric ester function, for preparation of monosaccharide building blocks leading to an iterative carbohydrate synthesis, (c) application of this methodology to the synthesis of a biologically important oligosaccharide.
