The programme comprises: (1) The choice of species for immunisation and of the source of antibodies. Currently most antivenom producers use horses but we have substantial evidence that favours sheep. We also intend to compare antibody production in other mammalian and avian systems. (2) Initial extraction and purification. These studies are designed to separate intact immunoglobulins of the IgG class from the many other proteins, especially albumin, found in whole blood, plasma, serum or the yolk of eggs. Such a step(s) will probably be the first stage in manufacturing a therapeutic product. (3) Final product purification and formulation. This comprises protease digestion of the antibodies; and Einal purification of the relevant immunoglobulin fragment. Final formulation will provide a stable Liquid dosage form. (4) Engineering the manufacturing system will aim to devise a simple and robust process that can be reliably implemented in developing countries that currently lack antivenom productio capabilities. Finally, (5), housing the manufacturing process will involve with engineering design companies to design a portable clean room facility for the entire manufacture of antivenoms.
