Fungal polyketide metabolites are a major potential source of novel bioactive compounds. Recent advances in bacterial polyketide biosynthesis have realised the goal of producing new compounds by rational manipulation of the biosynthetic pathway at the genetic level. In contrast, similar work on fungal polyketides, e.g. squalestatins and decarestrictines, will be isolated by synthesis of the potential advanced intermediates for biosyntheticstudies to elucidate the details of the assembly pathway and as the basis of a bioassay to allow the isolation and purification of the PKS enzymes for a reverse-genetics approach to isolate the PKS genes. A wide variety of modern synthetic methodology will be applied to the synthesis of the structurally complex intermediates. Assay procedures will make use of sensitive HPLC and HPLC/MS techniques. Partial amino acid sequence information from the purified enzymes will be used to design oligonucleotide probes to isolate the PKS for full sequencing.
