The central aim in modern biochemistry is to observe, as directly as possible, enzymatic processes in living cells. To achieve this, methods need to be developed for initiating reactions in very small subcellular volumes. Techniques of deep UV and multiphoton micro-irradation which will allow discrete areas of a cell nucleus to be irradiated offer a unique opportunity to study the dynamic responses of cell proteins to the induction of DNA damage. Once this is achieved the dynamic response, in terms of position, rate, topology and kinetics of DNA and nuclear proteins to UV-induced damage can be studied.Development of precision micro-masking and spectral tailoring of soft x-rays from a pulsed laser plasma source will allow the dynamics of intercellular communication between damaged and undamaged cells to be observed.This multidisciplinary project, at the boundary between the physical and life sciences, will provide state-of-the-art technology to underpin fundamental research in the areas of biochemistry and biological cell dynamics.
