This project is concerned with the synthesis of peptide analogues constrained to adopt parallel or anti-parallel beta-sheets. The constraint will be enforced by a norbornene unit and the work builds upon preliminary results which have proven the concept. The project will first investigate whether the norbornene constraints can impose beta-sheet formation on peptide sequences even if they do not contain amino acids which are prone to beta-sheet formation, and then determine how far along a peptide chain a single norbornene unit can induce beta-sheet formation. The modification of the norbornene residue to produce water soluble beta-sheets will be investigated as will the synthesis of more complex beta-sheet architectures comprising three or more peptide strands.
