This proposal aims to develop a new and original approach to the preparation of cyclobutanols. The approach will be stereoselective thus allowing complex molecules to be assembled rapidly with complete control of the product molecule's shape. Cyclobutanols are important organic structures which are found in biologically active molecules and are often used as intermediates for the synthesis of more complex compounds. A solid phase/combinatorial approach will be employed to speed up the usually slow process of developing a new synthetic reaction. The first total synthesis of the Pestalotiopsin natural products will then be used to illustrate the use of the new chemistry, and will allow the biological properties and pharmaceutical potential of these newly isolated compounds to be fully assessed.
