Both ROMPGELS and ROMPSPHERES will be applied in both parallel and combinatorial syntheses. ROMPGELS, which are insoluble polymers derived from the ring opening metathesis polymerisation of polyfunctional strained alkenes using the Grubbs catalyst C12 (Cy3P) 2Ru=CHPh, are useful for the rapid synthesis of target pharmaceuticals in the solution phase but with greatly simplified purification. These novel supported reagents will be adapted for acylation and Mitsunobu reactions, diverse carbon-carbon bond constructions and oxidation reactions. ROMPSPHERES are high loading, large resin bead block copolymers derived from the coating ring opening metathesis polymerisation of polyfunctional strained alkenes starting from polystyrene resin beads lightly functionalised with ArCH=RuC12 (Cy3P) 2 residues (100-200 mesh, 0.06 mequiv.g-1). ROMPSPHERES have the potential for use in diverse chemical transformations of solid supported substrates with high bead loading and with high-mass recovery per bead. This should greatly simplify single bead per well bioassays following cleavage from the support and where necessary full compound authentication without recourse to tagging or deconvolution by resynthesis. The products from both ROMPGELS and ROMPSPHERES syntheses will be used to produce diverse sub-libraries of compounds for bioassay.
