In a series of preliminary studies, most of which are unpublished, we have developed successful methods for the asymmetric transfer hydrogenation reactions of ketone substrates containing adjacent alkoxy and amino groups. This has been made successful through the selection of the appropriate protecting groups and represents a breakthrough in terms of versatility of this valuable synthetic process. The catalysts are comprised of ruthenium complexes of either an amino alcohol or monotosylated diamine.This application seeks funding to support a series of investigations designed to define the versatility and limitations of the new transformations which have been made possible by our developments in this area. The applications have been selected on the basis of their perceived novelty and synthetic value. This project is aimed principally at the applications of the methodology rther than investigations of its mechanism (which will be the subject of another application).In summary we intend to systematically examine the reduction of a,b, and g-alkoxy and amino substituted ketones, heterocyclic substrates and, in the case of the reversible amino alcohol system, in-situ trapping of kinetic products. Certain C=N reduction will also be studied.
