This proposal aims to provide a unique, highly efficient and rapid synthesis of the manzamine alkaloids. The chemistry involves, as its key step, a powerful and stereocontrolled intramolecular azomethine ylide cycloaddition. This process sets up two new rings and three new chiral centres in a single step, to provide, starting from easily-prepared ring A and ring E substrates, the ABCE rings of the manzamines. Ring D will be constructed from this tetracylce and the total synthesis completed in the shortest and most efficient route to date. The manzamines are an important class of marine alkaloids with significant biological activity as antileukemic and antibacterial agents.
