The objective is to develop a new method for studying DNA alkylation by drugs and carcinogens. We will develop reagents for efficient and selective mono-dealkylation of DNA. The method will depend on the transfer of an alkyl group from the phosphotriester to a cobalt (I) supernucleophile. We will study reactions of cob (I) alamin and cobaloxime (I) with nucleotides and DNA modified by alkylating drugs and carcinogens. The product alkylcobalamins will be analysed by HPLC and mass spectrometry, aided by isotopic labelling. The methodology may have important applications in cancer treatment and in monitoring human exposures to carcinogens.
