This project is concerned with the development of new linkers based on the perfluorosulfonate unit, for solid phase organic synthesis (SPOS). These can function as either 'traceless' linkers or as sites for the incorporation of additional diversity on cleavage of the product from the resin. The diversification step will be achieved by exploiting the known reactivity of triflates and nonaflates in transition metal coupling procedures exemplified by the Suzuki and Heck reactions. The project involves three basic stages. Synthesis of the perfluorosulfonic acids coupled to a suitable tethering unit and demonstration of the basic cleavage chemistry. Preparation of suitable solid supports containing the linker by either grafting to pre-formed polymer beads or by coupling to a monomer and in-house polymerisation. Verification of the SPOS chemistry and applications of these resins as both traceless linkers and as points for library diversification. In addition, application of the poly(perfluorosulfonic) acid polymer by-product as a resin capture agent for product purification will also be explored.
