A significant part of combinatorial chemistry is based on the reactions of supported organic entities with solution-based reagents. In some cases the reaction course nicely mirrors that seen in solution, but in others there are serious shortcomings. This limits the present power of solid-phase synthesis. There is surprisingly little quantitative work on comparing solution and supported-phase reactivity, which the present proposal attempts to rectify.The emphasis will be on reactions catalysed by metal complexes in homogeneous solution. There are two reasons for this; firstly, the proposer's experience and expertise lies in this field, and secondly, catalysis poses challenges whose solution will have more general applicability. This is because many catalytic cycles depend on the involvement of different reagents of varying structure, steric bulk and polarity at different points in the cycle for success. Solving the problems here is likely to provide guidelines of broad generality.The project will be facilitated by the synthesis of solid supports with defined side-chain structure and high levels of substitution.
