The large variety of intricate indole alkaloid skeleta have become proving-grounds for new synthetic methodologies. There is one skeletal type - indole 'alkaloids with a 7,16-bond' - for which no route has been described - akuammiline is typical. We propose developing a route to this structural type.Our scheme requires the evolution of two interrelated methodologies: (i) the conversion of pyrroles into 1-aza-1, 3-butadien-5-ones and (ii) the cycloaddition chemistry of such species. Some work has been reported on the cycloadditions of simpler 1-aza-1, 3-butadienes; there are no reports of 1-aza-1, 3-butadien-5-ones.We suggest that the routes and methodology which it is proposed to examine would be of value in heterocyclic synthesis in general and for heterocyclic natural product synthesis in particular. The proposed investigations would make an excellent area in which the training of a PhD synthetic organic chemist could take place.
