This proposal is concerned with new methods in both hardware (instrumentation) and software (image/signal processing) for use in thin tissue autoradiography. The motivation for this proposal is driven by the need to overcome the limitations of poor sensitivity and quantification associated with using conventional film emulsion. The resulting hardware and software techniques will then be used to explore novel autoradiographic imaging methods currently precluded because of the limitations of the available technology.Although previous work in this area has resulted in greatly enhanced sensitivity, these systems have generally exhibited poor resolution compared to film. However, direct detection with a Charge Coupled Device (CCD) has shown promising results (~60% detection efficiency, ~30% resolution) using common beta emitters (S35, P33) encountered in autoradiography. This proposal builds on this work by extending operation to higher energies (up to 2 MeV) and higher temperatures (~300k). In order to achieve this aim, novel pixel thresholding and particle tracking algorithms will be developed. The results of this work will be used in the design and construction of a hybrid CCD autoradiography device. Preliminary investigations into novel autoradiographic imaging methods will then be undertaken using the new imaging system.
