We will investigate the interaction between proteins and characterised biomaterial surfaces under physiological conditions. Such information is an essential step in the design of biomaterials (hyrogels, amphiphilic networks, polyurethanes ..) for specific applications within the body.Until now, a major obstacle to such work has been the heterogeneity of useful biomaterials, so that the protein adsoprtion characteristics vary from place to place on the biomaterial surface. Information on these adhesion characteristics is crucially important for predicting how biomaterials will behave as regards the activation of immune responses, the clotting cascade, and the performance of interocular lenses.We plan to overcome this obstacle by combining our existing techniques of (a) micro-thermal analysis, and (b) affinity mapping. We have developed technique (a) in order to combine imaging at sub-micron spatial resolution with chemical fingerprinting. Technique (b) maps the distribution and strength of intermolecular interactions over a sample.The resulting data will enable us to interpret findings on how biomaterial samples behave in clinical ophthalmic practice.
